Personalized Cancer Vaccines Show Promise for Melanoma

Cancer vaccines clinical trials cancer research treatment re-occurrence effectivity

"The number of patients who were treated is low, so there is certainly a likelihood that these patients would have stayed cancer-free without vaccine", said Ott.

"The promising results show that personalised cancer vaccines created to treat skin cancer are safe to use, and that for some patients, they can successfully harness the power of the body's own immune system to kill cancer cells", Pickworth added. It was tested in six people with melanoma who'd had their tumors removed by surgery.

Two companies reported that their personalized cancer vaccinations, created to target specific cancer mutations, could serve as viable add-ons to current immuno-oncology drugs in order to deliver a potent combination treatment to tumors.

Twelve skin cancer patients, out of a total of 19 across both the trials, avoided relapses for two years after receiving different vaccines developed by German and US teams, researchers reported in the journal Nature on Wednesday.

Antigens are molecules that are displayed on the surface of cells and stimulate the immune system. The vaccines were administered to patients to train their immune system to recognize these neoantigens, with the goal of stimulating the immune system to destroy the cancer cells that display them. Eight of them had detectable cancer post surgery.

European Union interior ministers vow support to Italy on migration
The service said one of its vessels intercepted a boat with 42 people aboard near the southwestern Trafalgar Cape late Wednesday. The interior ministers mentioned "disembarkation", issues in their final communique, but offered no details or commitments.

With swift shots to the arm, doctors safely and effectively prime our immune systems to fight off deadly infectious diseases.

Next-phase trials in larger groups of participants are now needed, he said, "to establish the efficacy of these vaccines".

A total of 13 patients were included in the first study and all of them had stage III or IV melanoma. Five additional patients had already started to experience tumor regrowth by the time of vaccination, but did appear to benefit as well, with tumors initially shrinking in two of them, and one patient experiencing complete remission after treatment with a PD-1 inhibitor. The results of the studies, however, show that researchers can now continue to develop new cancer vaccines. For their vaccines, the researchers used RNA fragments representing up to 10 neoantigens. Because some mutations are present in the DNA, but the gene is not made into RNA and protein, the researchers used RNA sequencing to identify mutations that caused the production of a mutated RNA, which is then normally translated into a protein. "These T cells discriminated mutated from wild-type antigens, and in some cases directly recognized autologous tumour". The study is considered as the world's first with a personalised mRNA vaccine. The vaccine was aimed at generating responses to the neoantigens from T cells of two kinds: CD8 killer cells and CD4 helper cells.

Experts plan to proceed to the second phase of clinical trials of vaccines, and to determine their effectiveness in combating other types of cancer. When the team monitored the vaccine's effects on the immune system in each patient, they found that both T-cell types had indeed been activated by the vaccine and could recognize the neoantigens bound to HLA molecules.
