"Jill and I were heartbroken to learn of the violent attack at a security checkpoint on the US Capitol grounds", Biden said in a statement.
In a statement, Pelosi said Congress was ready to assist law enforcement with the investigation into the vehicle attack.
In a tweet Friday, Senate Minority Leader Mitch McConnell said, "Praying for the United States Capitol Police officers who were attacked at the Capitol".
The authorities say he was not known to police and terrorism does not appear to have been a motive.
The incident occurred about 100 yards (91 meters) from the entrance of the building on the Senate side of the Capitol. One witness, the Rev. Patrick Mahoney, said he was finishing a Good Friday service nearby when he heard three shots ring out.

The Capitol complex has been on high alert since the January 6 attack by former President Donald Trump's supporters, which left five people dead.
President Joe Biden left Washington earlier in the day for Camp David, the presidential retreat in Maryland. Large sections of fencing that secluded much of the surrounding area for months have only begun to come down in recent weeks.
The FBI's Washington Field Office is responding to the situation and is now providing support to the Capitol Police.
There was a heavy police response outside of the Capitol, including a helicopter landing on the premises. "On behalf of the entire House, we are profoundly grateful".
There were reports of gun shots at the Capitol as the incident unfolded and police ordered staff via a broadcast text message to shelter in place.
A Sputnik correspondent at the scene witnessed and took photos of National Guard troops positioned around the perimeter, some carrying riot shields and other protective equipment.
Television footage showed a blue sedan crashed into a security barrier on one of the streets leading to Congress, as what appeared to be the injured officers were loaded onto gurneys and into ambulances. Law enforcement and paramedics could be seen caring for at least one unidentified individual.
